Once you have finished the sign-up process, you will be redirected to your download Book page.The major objectives of this project are: 1) to develop and define a method of increasing the phagocytosis of Escherichia coli and other gram negative bacteria by human peripheral blood leukocytes (PBL) and to isolate the effector cells and mechanism(s) involved, 2) to develop a model for in vitro study of phagocytosis and activation of the PBL and 3) to investigate the role of polymorphonuclear leukocytes (PMN) in an immune response. Phagocytosis of complement-opsonized zymosan particles has been found to involve a) PMN as the effector cells, b) ingestion of fluid-phase particles by PMN as a crucial event, c) the PMN G-protein-coupled receptors, a class of "signal transducing" receptors, are involved, d) the complement receptor, CR3, is also involved, e) phorbol myristate acetate (PMA) synergistically enhances the ingestion of complement-opsonized zymosan by the PMN, f) serotonin (5-HT) but not melatonin inhibits the ingestion of complement-opsonized zymosan by the PMN. To study these effects, freshly isolated human PMN have been used and activation is monitored by measurement of superoxide anion production and by differential cell counts.
